Applications
HAISIL HL “high load” columns and car-tridges are intended for applications requiring maximum loading and selectivi-ty. The high surface area permits high bonded phase loading yet the monofunc-tional bonded phase and the absence of micro-pores eliminates the problems that plague many of the so-called “high capac-ity” columns currently on the market. The Type B silica backbone is pure, robust, and stable under harsh condi-tions. Users repor t satisfactory lifetime when used for prolong periods at pH as high as 9.
High Speed Analysis of Paraben Food Preservatives
Parabens represent another example of food additives that are routinely analyzed by HPLC. Often, the food matrix being assayed for preservative content requires exhaustive cleanup and pre-filtration in order to prevent clogging of columns with nonporous or 2µm sorbents recently suggested for this analysis. As in the previous diet cola example, the compounds of interest are base line resolved under isocratic conditions. The inexpensive HAISIL HL 5µm 20x2.1mm cartridge column and the 50 second analysis time contribute significantly towards reducing operating costs.

Abstract
Previously, we presented a complete 20-minute "fast" Edman chemistry cycle for the Applied Biosystems Protein Sequencer, HT series (49X-HT). Here we describe a new "fast" cycle for the high sensitivity cLC series (49X-cLC) instrument.The 35-min cycle includes a 20-min reversed phase capillary HPLC separation of the phenylthiohydantoin (PTH)-amino acids.The normal cycle time of the 49x-cLC was approx. 50 minutes; therefore approximately 10 hours were required to complete 10 residues of sequencing.With the new cycle, 10 residues of sequencing can be
completed in only 7.5 hours. Using this rapid cycle on the 492 or 4-cLC protein sequencer, 10 residues of sequencing on 2 different proteins can be automatically completed within a night.
In developing this rapid cycle, we have created new car tridge and flask cycles and sequencing methods.We are using a shor ter C18 RP-HPLC capillary column for the chromatography and have devel-oped a new gradient. We will describe all the changes made to the standard system.We will also present protein and peptide sequencing results that were generated using this rapid cycle.
